How to inhibit a protease that cuts the induced protein?
We have been trying to induce a mutated protein. The wild type was purified. But while induction of this new protein it was seen that Asp-N-endopeptidase cleaved the protein. How to prevent this protease action so that the protein could be purified?
What expression system are you using? If you are using E. coli, a change to a different host might help.
Addition of protease inhibitor cocktail might help.
